how would you respond if you think that someone is flirting with your date,spouse?

 I think it would depend on how my date/spouse reacts to that flirting. If she ignores it and is into the setting we have created, then no harm done. I might even make a point to tease her and say, "Hey, old boy is checking you out. Wanna make it a threesome?" Ha. Well, maybe not the threesome--yet.

If attentions are divided, I'd get up and beat the shit out of him, or her, for flirting with my date. Then I'd ask my date if she is with me or with him, and that I don't appreciate that she is entertaining someone else's overtures if she's supposed to be with me. Otherwise, just say the word and I'm ghost. More fish in the sea, you know.

It is all a question of degree. Also depends on the individuals involved. We all don't process the same so the response will most definitely vary.
